It is probably because the copper is not tinned. This means it will have a thin coating of tarnish on top which acts as an insulator and that is what gives your meter problems. If you scratch the tip of the probe on the copper you will get a better reading. There is nothing wrong with the prototype baord.

When I use that type of protoboard, I give the copper a quick once over with a "ScotchBrite" type abrasive pad to make it nice and shiny before soldering the components in.
