Hi, I would like to print my first PCB design that I made with Sprint Layout 6 and I have a question: According to the manufacturer the min track/spacing for low coast PCB is 6/6 mil.

Does it means that if I'm working on a grid of 158.75 um (6.25mil) the black lines between the green tracks and the space between one track to the other should be more of one grid division?

Seems so. 0.16mm trace width / spacing can get you 30 pcb traces in parallel in between the body ends of a single 1/3W thru hole resistor ... Not a home etch job.
 
6/6 definitely means 6 thou tracks and 6 thou gaps. I remember back in the 80s 12 thou track and 12 thou gap was state of the art but I still prefer to use 20 thou tracks for audio.
